In this model, LLC-PK1 cells are cultured on Transwell inserts until they form a confluent, polarized monolayer with functional tight junctions. Monolayer integrity is verified e.g., by measuring transepithelial electrical resistance (TEER) or the permeability of Lucifer Yellow before transport experiments.<\/p><p>For permeability studies, the test compound is added to either the apical or basolateral compartment, and bidirectional transport (apical-to-basolateral and basolateral-to-apical) is measured over an incubation period. The apparent permeability coefficient is calculated from the rate of compound transport across the membrane, while the efflux ratio is used to identify transporter-mediated efflux. Drug recovery is also determined to detect intracellular accumulation.
