{"data":{"id":72,"name":"Calu-3 Permeability Assay","abbreviation":"CALU-3_assay","description":"<p>The Calu-3 permeability assay is an <em>in vitro<\/em> model used to evaluate the barrier function and permeability of the human airway epithelium. Calu-3 cells, derived from human bronchial submucosal glands, are cultured on permeable Transwell inserts at an air\u2013liquid interface until they form a confluent, polarized monolayer with tight junctions that closely resembles the bronchial epithelial barrier.<\/p><p>The integrity of the epithelial barrier is assessed by measuring transepithelial electrical resistance (TEER), which correlates with the permeability of FITC-dextran across the monolayer. A decrease in TEER indicates increased epithelial permeability and disruption of tight junctions. During experiments, Calu-3 monolayers are exposed to different environmental conditions, including varying oxygen concentrations, positive airway pressure, or pharmacological agents. Changes in barrier function are evaluated by monitoring TEER together with the flux of marker molecules, inflammatory cytokine secretion, total protein release, cell viability, and histological morphology.
