Exchange between internal and external water pools alters the linewidth of the \u00b9\u2077O NMR signal of external water, enabling calculation of permeability coefficients. The approach is highly sensitive to rapid water exchange, making it suitable for polyunsaturated lipid bilayers that exhibit high permeability. Results demonstrated that increased acyl chain unsaturation enhances water transport due to looser lipid packing, while ethanol or incorporation of phosphatidylethanolamine decreases permeability by tightening bilayer packing.
