Bio-mimetic Artificial Membrane Permeation Assay (BAMPA) is a permeability assay developed to predict passive intestinal drug absorption. It is an improved version of the Parallel Artificial Membrane Permeation Assay (PAMPA), in which the artificial membrane is modified to better mimic the lipid composition of the intestinal brush border membrane. Unlike cell-based assays, BAMPA contains no living cells, pores, or active transporters and therefore measures only passive transcellular diffusion.

The assay uses a donor and an acceptor compartment separated by a porous filter impregnated with a biomimetic lipid solution mimics intestinal brush border membrane. The test compound is added to the donor side and diffuses across the artificial membrane into the acceptor compartment. After incubation, the compound concentration in the acceptor chamber is quantified to calculate the apparent membrane permeability. The optimized membrane composition provides a closer approximation to intestinal membrane properties and improves the prediction of human oral absorption. BAMPA is widely used as a rapid, inexpensive, and high-throughput screening method during early drug discovery for evaluating passive permeability
