The Ussing chamber is a classical ex vivo technique for studying transport and barrier properties of epithelial tissues or cell monolayers. In this setup, a tissue or monolayer is mounted between two fluid-filled chambers, isolating the apical and basolateral sides. Electrodes measure potential difference and current, allowing determination of transepithelial resistance (TEER) and short-circuit current as indicators of ion flux and barrier integrity. Meanwhile, tracer molecules (e.g. fluorescein, FITC-dextran) can be applied to one side and quantified in the opposite chamber to derive flux and permeability coefficients. The method enables separation of transcellular and paracellular transport, assessment of active transport, and study of tight-junction dynamics. Ussing chamber remains a gold-standard tool in epithelial physiology, pharmacology, and intestinal permeability research.

source of image: 10.1007/978-3-319-16104-4_24 (Chapter: Ussing Chamber)
