The parallel artificial membrane permeability assay (PAMPA) is a system designed to quantify passive diffusion using a simple two-compartment setup. The assay consists of a donor and an acceptor well separated by a microfilter impregnated with a lipid solution that mimics the hydrophobic core of biological membranes . The artificial membrane is prepared by depositing a defined amount of phospholipid or lipid mixture dissolved in an organic solvent onto the porous filter support, creating a stable and reproducible barrier suitable for high-throughput analysis. Test compounds are placed in the donor compartment, and their passive permeation is determined by quantifying their appearance in the acceptor compartment after incubation. Because no living cells or extended culture times are needed, PAMPA enables rapid parallel screening of large compound sets, in contrast to CACO-2 assays. Specialized variants, such as skin-PAMPA, modify membrane composition—e.g., ceramides, fatty acids, and cholesterol—to model barrier-specific lipid environments for improved physiological relevance.

source of image: 10.3390/mi12030294
