Binary mixtures of (egg-derived) phosphatidylcholine (PC) and phosphatidic acid (PA) combine a neutral, fluid bilayer matrix with negative surface charge, enabling studies of electrostatics and permeability. The PC provides a loosely packed, hydrated platform, while PA—bearing a small, anionic headgroup—imposes charge and local curvature stress. Inclusion of PA influences lipid packing, promotes interleaflet coupling, and can accelerate transbilayer lipid movement under perturbation (e.g. conversion of PC to PA by phospholipase D induces rapid lipid scrambling). Such charged membranes serve as simplified models to probe how membrane charge and packing modulate solute partitioning, ion diffusion, and barrier properties.
