Calu-3 is a human bronchial submucosal gland carcinoma cell line widely used as an in vitro airway epithelial model. When grown at an air–liquid interface, Calu-3 cells form polarized monolayers with tight junctions, high transepithelial resistance, and mucus secretion, resembling the conducting airway epithelium. They express functional ion transport systems, including CFTR-mediated chloride and bicarbonate secretion, and serve as a model of submucosal gland serous cell activity.

Calu-3 cells also express uptake and efflux transporters relevant to pulmonary drug disposition, including P-glycoprotein and BCRP. Their secretory activity, barrier properties, and inflammatory responses make them valuable for studying respiratory drug delivery, infection, airway inflammation, and injury induced by oxygen or mechanical ventilation.

Cellosaurus: https://www.cellosaurus.org/CVCL_0609
