A hexadecane liquid layer is widely used as an artificial membrane to model passive transcellular diffusion in permeability assays (e.g. HDM-PAMPA). A thin (≈9–10 µm) hexadecane layer separates donor and acceptor compartments, providing a reproducible hydrophobic barrier. It mimics the hydrocarbon core of phospholipid bilayers, which represents the principal barrier to passive diffusion in biological membranes.

Permeability values obtained correlate well with human gastrointestinal absorption and offer better predictive power than simple partition coefficients. The system also enables pH-dependent profiling and correction for unstirred water layer effects. While it does not capture active transport or efflux mechanisms, the hexadecane membrane provides a simple, high-throughput, and robust tool for ranking compounds by passive permeability in early drug discovery.
